Pleiotropy

Introduction
Pleiotropy is a phenomenon where a single gene influences multiple, seemingly unrelated phenotypic traits. This occurs because the gene product is involved in multiple developmental or physiological processes.
Core concept
Definition: one gene → multiple phenotypic effects. Mechanism: the protein product of the gene functions in multiple tissues, developmental pathways, or metabolic processes. Examples: Phenylketonuria (PKU): mutation in PAH gene (phenylalanine hydroxylase). Primary effect: cannot convert phenylalanine → tyrosine. Pleiotropic effects: intellectual disability (phenylalanine toxicity in brain), lighter skin/hair (less tyrosine → less melanin), musty body odour (phenylacetic acid accumulation), eczema, seizures. All from one gene mutation. Sickle cell anaemia: β-globin mutation → HbS → sickle cells → pleiotropic: haemolytic anaemia, vaso-occlusion (pain crises, organ damage), stroke, splenic sequestration, increased Pneumococcus susceptibility. Marfan syndrome: FBN1 gene (fibrillin-1). Affects connective tissue throughout body → tall stature, long fingers (arachnodactyly), aortic aneurysm, lens dislocation, flexible joints.
Go deeper
Pleiotropy vs polygenic: Pleiotropy: 1 gene → many traits. Polygenic: many genes → 1 trait. Both are important modifications of simple Mendelian genetics. A gene may be both pleiotropic (affecting multiple traits) and part of a polygenic system (one of many genes affecting one trait). Housekeeping genes: expressed in virtually all cell types → mutations in them tend to have widespread pleiotropic effects → often lethal when homozygous.
From Class 10
Usually, one gene affects one trait. But in pleiotropy, a single gene affects multiple traits simultaneously — like a switch that controls several functions at once.
