Mutation and Aberrations

Introduction

Mutations are heritable changes in the genetic material — ranging from point mutations (single nucleotide changes) to chromosomal aberrations (large-scale structural or numerical changes in chromosomes) — and are the ultimate source of all genetic variation.

Core concept

Point mutations (gene mutations): substitution: one base replaced by another. Transition: purine ↔ purine or pyrimidine ↔ pyrimidine (A↔G, C↔T). Transversion: purine ↔ pyrimidine. Effects: silent/synonymous (same amino acid — degeneracy of code), missense (different amino acid — may alter function), nonsense (creates stop codon → truncated protein). Insertion/deletion (indels): one or more bases inserted or deleted → frameshift mutation → all downstream codons change → usually loss of function. Sickle cell anaemia = point mutation (missense): GAG → GTG (Glu → Val). PKU = point mutation in PAH gene. Chromosomal aberrations: Structural: Deletion (loss of segment), Duplication (extra copy of segment), Inversion (segment reversed), Translocation (segment moves to non-homologous chromosome). Numerical (aneuploidy): Nondisjunction during meiosis → monosomy (2n-1) or trisomy (2n+1). Polyploidy: extra complete chromosome sets (3n, 4n) — common in plants. Causes of mutation: spontaneous (errors in DNA replication). Induced: mutagens (UV, X-rays, chemical mutagens like alkylating agents, base analogues, intercalating agents).

Go deeper

Frameshift mutations and consequences: single base insertion or deletion → shifts the reading frame by 1 or 2 → all downstream codons changed → usually nonsense codons encountered early → truncated, non-functional protein. Insertion or deletion of 3 bases → in-frame → adds/removes one amino acid → less disruptive (if not critical domain). Example: cystic fibrosis most common mutation (ΔF508): deletion of 3 bases → loss of phenylalanine at position 508 → CFTR protein misfolded → degraded.

From Class 10

A mutation is a permanent change in the DNA sequence of a gene. Mutations can be as small as a single base change or as large as a rearrangement of whole chromosomes.
