Transcription Process
Introduction
Transcription is the copying of genetic information from one strand of DNA into RNA. Only a segment of DNA (a transcription unit) and only one strand is copied.
Core concept
Transcription unit: promoter (towards the 5' end of the coding strand, binds RNA polymerase), structural gene, terminator. Template strand has 3'→5' polarity; the coding strand has the same sequence as the RNA (with T instead of U) and is not copied. Bacteria: one DNA-dependent RNA polymerase; initiation factor sigma (σ) for initiation, termination factor rho (ρ) for termination; mRNA needs no processing and translation can begin before transcription ends. Eukaryotes: RNA polymerase I → rRNAs (28S, 18S, 5.8S); II → hnRNA (precursor of mRNA); III → tRNA, 5S rRNA, snRNAs. hnRNA processing: splicing (introns removed, exons joined), capping (methyl guanosine triphosphate added to the 5' end) and tailing (poly-A tail of 200-300 adenylates at the 3' end).
Go deeper
Split genes (exons and introns) are a feature of eukaryotes; spliceosomes that remove introns are therefore not found in prokaryotic cells. Introns are thought to be a remnant of an ancient past.
From Class 10
A gene is like a recipe in a cookbook; transcription is photocopying just that one recipe (as RNA) to take into the kitchen.
