Semiconservative Replication
Introduction
DNA replication is semiconservative: each new DNA molecule has one parental (old) strand and one newly synthesised strand, as Watson and Crick predicted.
Core concept
Proof: Meselson and Stahl (1958) grew E. coli in 15NH4Cl (heavy nitrogen) for many generations, then moved them to 14NH4Cl. DNA was separated by caesium chloride (CsCl) density-gradient centrifugation. After one generation (20 minutes): all DNA hybrid (intermediate density). After two generations (40 minutes): equal amounts of hybrid and light DNA. Taylor and colleagues later showed the same in Vicia faba (faba beans) using radioactive thymidine. Machinery: replication starts at the origin; DNA-dependent DNA polymerase adds nucleotides only in the 5'→3' direction; deoxyribonucleoside triphosphates are both substrate and energy source. The leading strand is made continuously; the lagging strand is made discontinuously as Okazaki fragments that are joined by DNA ligase. In eukaryotes replication occurs in the S phase.
Go deeper
Because polymerase works only 5'→3', the two strands at the replication fork are copied differently — continuously on one, in fragments on the other. In E. coli, about 4.6 × 10^6 bp are copied in roughly 18 minutes, so the polymerase is extremely fast and accurate.
From Class 10
Before a cell divides, its DNA must be copied so each daughter cell gets a full set — that copying is replication.
