Chemiosmosis in Chloroplasts
Introduction
Chemiosmosis in chloroplasts is the mechanism of ATP synthesis during light reactions, where a proton (H⁺) gradient across the thylakoid membrane drives ATP synthase (CF₁-CF₀ complex) to synthesize ATP.
Core concept
H⁺ accumulates inside thylakoid lumen by: (1) water splitting at PSII releases H⁺ into lumen, (2) plastoquinone (PQ) carries H⁺ from stroma to lumen during electron transfer. Proton gradient: high H⁺ inside thylakoid lumen, low H⁺ in stroma. H⁺ flows from lumen to stroma through ATP synthase (CF₀-CF₁ complex) → conformational change → ADP + Pᵢ → ATP. This is photophosphorylation.
Go deeper
Mitchell's chemiosmotic hypothesis (1961, Nobel 1978): ATP synthesis is driven by proton motive force (PMF) = membrane potential + pH gradient. Same principle in chloroplasts and mitochondria, but opposite direction: in mitochondria H⁺ is pumped out (into IMS), in chloroplasts H⁺ is pumped in (into thylakoid lumen).
From Class 10
In mitochondria, ATP is made by chemiosmosis — proton gradient driving ATP synthase. The same principle works in chloroplasts during light reactions.
