Restriction Endonucleases
Introduction
Restriction endonucleases are 'molecular scissors' — enzymes that cut DNA at specific recognition sequences. They are the basic tool of genetic engineering.
Core concept
In 1963 two enzymes restricting the growth of bacteriophage in E. coli were isolated: one added methyl groups to DNA, the other cut DNA (restriction endonuclease). The first restriction endonuclease was Hind II, which always cuts DNA at a particular point by recognising a specific sequence of six base pairs. Today more than 900 restriction enzymes are known, from over 230 strains of bacteria. Naming: EcoRI — E (genus Escherichia), co (species coli), R (strain RY 13), I (first enzyme isolated from that strain). Nucleases are of two kinds: exonucleases remove nucleotides from the ends of DNA; endonucleases make cuts at specific positions within the DNA. Each restriction endonuclease recognises a specific palindromic nucleotide sequence and cuts each of the two strands of the double helix at specific points in their sugar-phosphate backbones (phosphodiester bonds). Many enzymes cut a little away from the centre of the palindrome, between the same two bases on opposite strands, leaving single-stranded overhangs called sticky ends, which form hydrogen bonds with complementary cut counterparts; DNA ligase then joins them.
Go deeper
Sticky ends are what make recombinant DNA possible: cutting the vector and the source DNA with the same enzyme gives matching ends that can be joined.
From Class 10
Just as scissors cut paper at a marked line, restriction enzymes cut DNA only at particular letter sequences.
