Pyruvate Dehydrogenase Complex

பைருவேட் டீஹைட்ரஜனேஸ் காம்ப்ளக்ஸ்

NCERT Class 11 · Respiration in Plants · 0 NEET questions

Glycolysis ends in the cytosol, but the Krebs cycle demands Acetyl CoA in the mitochondrial matrix. The Pyruvate Dehydrogenase Complex is the mandatory gatekeeper. Miss this link, and cellular respiration halts entirely.

The idea

This multi-enzyme complex catalyzes the oxidative decarboxylation of pyruvic acid. It converts one molecule of pyruvate into one molecule of Acetyl CoA, one NADH, and one CO2. The reaction requires NAD+ and Coenzyme A as essential cofactors. It occurs in the mitochondrial matrix, bridging the anaerobic glycolysis and the aerobic Krebs cycle.

Why it matters

It exists to convert the three-carbon pyruvate into the two-carbon acetyl group, which is the only form that can enter the mitochondrial TCA cycle.

NEET lens

NEET focuses on the products: 1 Pyruvate yields 1 Acetyl CoA, 1 NADH, and 1 CO2. Remember, this reaction occurs twice per glucose molecule. Do not confuse the location; it is strictly mitochondrial matrix. Watch for questions asking for the 'link reaction'—this is it.

Clinical significance

Deficiency in the E1 subunit of this complex leads to Pyruvate Dehydrogenase Deficiency, causing severe lactic acidosis. The brain, being highly dependent on aerobic respiration, suffers irreversible neurological damage. Without this complex, cells shift to anaerobic metabolism, which is insufficient for human survival.

Introduction

The pyruvate dehydrogenase complex (PDC) is a large multienzyme complex in the mitochondrial matrix that converts pyruvate to acetyl-CoA, linking glycolysis to the TCA cycle.

Core concept

Reaction: Pyruvate (3C) + CoA + NAD⁺ → Acetyl-CoA (2C) + CO₂ + NADH. Location: mitochondrial matrix. This is oxidative decarboxylation (pyruvate is oxidized and loses CO₂). Three enzymes in complex: E1 (pyruvate decarboxylase, needs TPP/thiamine), E2 (dihydrolipoyl transacetylase, needs lipoic acid and CoA), E3 (dihydrolipoyl dehydrogenase, needs FAD and NAD⁺). Products: 1 acetyl-CoA + 1 CO₂ + 1 NADH per pyruvate. Per glucose: 2 acetyl-CoA + 2 CO₂ + 2 NADH.

Go deeper

PDC regulation: inhibited by acetyl-CoA (product), NADH (product), ATP (high energy signal) — feedback inhibition. Activated by CoA, NAD⁺, AMP/ADP (low energy). PDC kinase phosphorylates PDC → inactive. PDC phosphatase dephosphorylates → active. This tightly controls carbon flow into TCA cycle.

From Class 10

Pyruvate from glycolysis must be converted before entering the Krebs cycle. The pyruvate dehydrogenase complex (PDC) performs this critical conversion.
