DNA Packaging
DNA பேக்கேஜிங்
A human cell contains two meters of DNA, yet it fits inside a nucleus measuring mere micrometers. If you think this is magic, you will fail NEET. This is pure structural engineering at the molecular level.
The idea
DNA is negatively charged due to phosphate groups and wraps around a positively charged histone octamer. This complex is the nucleosome. These nucleosomes repeat to form 'beads-on-a-string' chromatin, which further coils into highly condensed chromosomes during cell division.
Why it matters
DNA is extremely long (approx. 2.2 meters in humans) and must be tightly packed to fit within the microscopic nucleus of a cell.
NEET lens
NEET focuses on the charge of histones (positive) and DNA (negative). Remember: Histones are rich in basic amino acids, Lysine and Arginine. Do not confuse 'Euchromatin' with 'Heterochromatin' regarding their staining and transcriptional activity.
Clinical significance
Epigenetic regulation relies on histone modification. If DNA packaging fails, gene expression is disrupted, leading to oncogenesis or developmental disorders. Proper chromatin remodeling is essential for cellular homeostasis.
Introduction
A human cell holds about 2.2 m of DNA in a nucleus only about 10^-6 m across. It fits because DNA is packaged around proteins into chromatin.
Core concept
Prokaryotes: no defined nucleus; negatively charged DNA is held with positively charged proteins in a region called the nucleoid, in large loops. Eukaryotes: histones are positively charged basic proteins rich in lysine and arginine. Eight histone molecules form a histone octamer; negatively charged DNA wraps around it to form a nucleosome, which contains about 200 bp of DNA helix. Nucleosomes form 'beads-on-a-string' chromatin seen under the electron microscope, which coils further into chromatin fibres and, at metaphase, chromosomes. Higher-level packaging needs non-histone chromosomal (NHC) proteins. Euchromatin: loosely packed, stains light, transcriptionally active. Heterochromatin: densely packed, stains dark, inactive.
Go deeper
Numerical: length of DNA = bp × 0.34 nm. For E. coli (4.6 × 10^6 bp) that is about 1.36 mm. If a cell has 6.6 × 10^9 bp, total length ≈ 2.2 m. Histone H1 binds linker DNA between nucleosomes and helps further condensation.
From Class 10
Imagine winding a very long thread around many small spools so it fits in a matchbox — that is how DNA is packed.
