Protein Structure
புரத அமைப்பு
Biology is not a collection of shapes; it is a hierarchy of folding. If a protein misfolds, the cell dies. NEET examiners love to test if you understand that primary structure dictates the final 3D conformation.
The idea
Proteins are heteropolymers of amino acids. The primary structure is the linear sequence of amino acids. Secondary structure involves folding into alpha-helices or beta-pleated sheets via hydrogen bonds. Tertiary structure represents the complex 3D folding of the polypeptide chain, while quaternary structure describes the arrangement of multiple polypeptide subunits.
Why it matters
The specific 3D conformation of a protein is critical for its biological activity and functional specificity.
NEET lens
NEET focuses on the types of bonds stabilizing each level. Remember: Primary uses peptide bonds, Secondary uses hydrogen bonds, and Tertiary uses disulfide, ionic, and hydrophobic interactions. Do not confuse the levels; questions often ask which structure is essential for enzyme activity. Focus on the 3D folding requirement for functional proteins.
Clinical significance
Prion diseases like Creutzfeldt-Jakob disease occur due to protein misfolding. When the tertiary structure of a protein is disrupted, it loses its biological activity, a process known as denaturation. This loss of structure renders enzymes non-functional, leading to metabolic failure.
Introduction
Protein structure is organized at four levels: primary (amino acid sequence), secondary (local folding), tertiary (overall 3D shape), and quaternary (multiple subunits).
Core concept
Primary: specific sequence of amino acids, determined by DNA. Secondary: α-helix (H bonds between backbone NH and C=O, 3.6 aa per turn) and β-pleated sheet (H bonds between parallel strands). Tertiary: overall 3D structure (disulfide bonds, ionic bonds, hydrophobic interactions, H bonds with R groups). Quaternary: association of multiple polypeptide chains (subunits).
Go deeper
Chaperone proteins (Hsp70, GroEL) help newly synthesized proteins fold correctly by preventing premature aggregation. Without chaperones, many proteins misfold, especially under stress.
From Class 10
Amino acids link together in chains to make proteins. These chains fold into specific shapes. Protein structure has four levels.
